WASHINGTON (AP) ? In campaign mode for months, President Barack Obama is making his quest for a second term official with rallies in Ohio and Virginia while casting Republican rival Mitt Romney as a flip-flopping protector of the rich.
The events Saturday at two universities, Ohio State and Virginia Commonwealth, were billed as the official kickoff of Obama's re-election bid, even though he's been solidly engaged in his campaign and over a year ago filed the necessary paperwork to run again.
During the events, the president planned to try to convince voters that his policies have put the economy on more solid footing despite fresh evidence that the job market remains weak. He also was expected to try to define Romney as a candidate peddling failed policies for both the economy and national security.
Obama has headlined dozens of fundraisers around the country as his campaign tries to build a solid money advantage over Romney. In his official White House travels, often to the most contested states, the president has pitched policy positions that fit neatly into the campaign's central theme of economic fairness. They range from a millionaires' tax to freezing student loan interest rates.
Official campaign rallies can free Obama up to take more direct aim at Romney. Until now, Obama has used Romney's name sparingly, often choosing instead to cloak his criticisms of Romney in attacks against generic Republicans.
Some Democrats saw Saturday's rallies as a chance for Obama to put Republicans on notice that he plans to be an aggressor in the race.
"What we've seen too many times in the past is Democrats are way too meek in defining their opponents or defining themselves in an election," said Maria Cardona, a Democratic strategist. "This president is not going to let the Republicans define him."
Obama's speeches Saturday weren't expected to differ greatly from what he's been saying in fundraisers or what his team has said in the campaign.
David Axelrod, an Obama senior adviser, said the president wasn't a candidate who "reinvents himself week to week," as Axelrod poked at Romney's sometimes shifting positions. Instead, Axelrod and other advisers Obama would reinforce broader themes of advocating for the middle class and trying to portray Romney as the candidate for the wealthiest Americans.
Republicans argue the Obama campaign is not aiming for consistency, but rather struggling to find a comprehensive vision for a second term.
"They have nothing positive to run," said Sean Spicer, communications director for the Republican National Committee. "No successful incumbent, no impressive record and no thriving economy."
The latest job numbers from Friday highlighted the challenge Obama faces in convincing voters that he is the right steward for the economy. Job growth slumped for a second straight month. The unemployment rate dropped to 8.1 percent but largely because more people stopped looking for work and therefore were no longer deemed unemployed.
In the face of continued economic unease, Obama's rallies Saturday's were intended to recapture some of the youthful, hopeful energy of his 2008 campaign.
The campus settings were likely to create the atmosphere where Obama is at his best, feeding off the energy of an enthusiastic crowd. Young voters were a crucial voting bloc in 2008 victory.
Michelle Obama was joining the president at the stops in states critical to the re-election effort.
In 2008, Obama won Ohio while reversing decades of Republican dominance in Virginia.
Since then, Virginia has swung back toward the GOP in statewide elections. Both Obama and Romney advisers acknowledge that the state is up for grabs.
During a campaign event in Portsmouth, Va., on Thursday, Romney said, "This may well be the state that decides who the next president is."

Scientists identify prostate cancer stem cells among low-PSA cells
ScienceDaily (May 4, 2012) ? Prostate cancer cells that defy treatment and display heightened tumor-generating capacity can be identified by levels of prostate specific antigen (PSA) expressed in the tumor cells, a research team led by scientists at The University of Texas MD Anderson Cancer Center reports in the May 3 edition of Cell Stem Cell.
"Using a new technique, we were able for the first time to separate low-PSA and high-PSA prostate cancer cells. This led to the discovery of a low-PSA population of cancer stem cells that appears to be an important source of castration-resistant prostate cancer," said study senior author Dean Tang, Ph.D., professor in MD Anderson's Department of Molecular Carcinogenesis.
Hormone therapy is used to block production of testosterone, which fuels prostate cancer growth, via either chemical or physical castration. Tumors eventually resist this approach.
In cell lines and mouse model experiments, the low-PSA cells resisted chemotherapy and thrived under hormone deprivation, the two main prostate cancer drug treatments , the researchers found.
Low-PSA cells were found to be both self-renewing and capable of differentiating into other prostate cancer cell types upon division, a hallmark of stem cells called asymmetric cell division.
"Asymmetric cell division is the gold standard feature of normal stem cells," Tang said. "Using time-lapse fluorescent microscopy, we were able to show asymmetric cell division by filming a low-PSA cell dividing into one high-PSA cell and one low-PSA cell."
Their findings point to the need to develop new therapeutics to target low-PSA prostate cancer cells that can be combined with hormone therapy to wipe out cancer cells and prevent recurrence.
Low-PSA tumors associated with advanced prostate cancer
Previous research by others indicated that low-PSA tumor cells are rare in early stage disease but become more abundant in advanced prostate cancer. And patients whose tumors were composed of more than 50 percent PSA-positive cells enjoyed longer survival.
This made Tang and colleagues wonder whether the two cell types fundamentally differ from each other and so play different roles in prostate cancer progression.
They analyzed 20 untreated prostate cancer samples staged at varying degrees of aggressiveness on the Gleason scale and 23 samples where treatment had failed. High-PSA tumor cells composed on average about 80 percent of the Gleason 7 tumors, about 60 percent of the higher grade Gleason 9/10 tumors but only about 15 percent of the treatment-failed tumors.
A database analysis of gene expression in prostate cancer was consistent with the tumor experiments, indicating an association between low levels of PSA in tumors and disease recurrence, spread to lymph nodes, metastasis and shortened patient survival.
Turning high-PSA cells green
The next challenge was separating the two cell types to understand their biological, tumor-initiating and drug-response properties.
Tang and colleagues developed a lentiviral delivery system equipped with a PSA promoter that triggers green fluorescence protein when PSA is expressed in an infected cell. This lentiviral reporter system allowed separation of low-PSA and high-PSA cells for the first time, Tang said. A series of experiments uncovered striking differences between them.
High-PSA prostate cancer cells:
? Divide rapidly, which makes them vulnerable to chemotherapy that targets fast-proliferating cells;
? Express high levels of the androgen receptor, a key to testosterone production, and so are vulnerable to hormonal therapy; and
? Produce only identical copies of themselves when they divide.
Low-PSA prostate cancer cells:
? Divide slowly and express anti-stress genes that help them resist chemotherapy;
? Either lack or have a weak presence of the androgen receptor, allowing them to grow while hormonal therapy wipes out PSA-positive cells; and
? Can divide into one copy of themselves and one PSA-positive cell during reproduction. The researchers captured this on video microscopy, filming the division of a grey low-PSA cell into one copy of itself and one copy of a vibrantly green PSA-positive cell.
Low-PSA tumors have long-term tumor-generating capacity
When the team implanted the two cell types in hormonally intact male mice, the rapidly reproducing PSA-positive cells caused faster growth and larger tumors in the first generation. However, after that the low-PSA cells generated larger, faster-growing tumors and tumor incidence in the high-PSA cells dropped. In fact, the low-PSA prostate cancer cells possess indefinite tumor-propagating capacity.
In contrast, when implanted in the castrated mice, the low-PSA prostate cancer cells developed much larger tumors than the corresponding high-PSA cells. In another experiment, mice with tumors generated by either cell type were then castrated and treated with hormonal therapy. Low-PSA tumors grew better in these doubly androgen-deprived mice than the high-PSA tumors.
"These findings closely resemble progression observed in patients after androgen-deprivation treatment and reflect reducedPSA-producing cells in patient tumors after androgen depletion," Tang said.
Effect of low-PSA cells in human tumors
Tang and colleagues analyzed tumor cell PSA expression in 556 human tumors and found low protein levels correlated with reduced overall survival.
They separated the two types of cells in three primary human tumors and found that low-PSA cells did not express androgen receptor and have higher cell-generating and sphere-forming capabilities than high-PSA cells.
Future research will focus on developing therapeutic targets for low-PSA cells and illuminating the epigenetic landscapes of both cell types.

The Cabarrus County Sheriff's Office has identified the man in the video as 17-year-old Jonathan Alexander Wiles, and deputies reportedly spent more than an hour at his home after learning of the clip showing him firing shots at an anti-Amendment One campaign sign, <a href="http://www.wbtv.com/story/18018096/investigation-launched-after-video-ammendment-one-sign-shooting-goes-viral" target="_hplink">according to WBTV</a>. MINNEAPOLIS, M.N. -- "I believe that people who are either transsexuals or transvestites -- I don't know what the proper term is -- traditionally are more liberal-minded thinking people, tend to associate more with the defendants because, obviously, they have been either ridiculed before or are feeling in a position of being in a microscope all the time and are outcasts which lends themselves to associating more with the defendant." Methodists Maintain Homosexual Acts Are 'Incompatible With Christian Teaching'
Despite emotional protests and fierce lobbying from gay rights groups, United Methodists voted on Thursday (May 2) to maintain their denomination's stance that homosexuals acts are "incompatible with Christian teaching." Two "agree to disagree" proposals were soundly defeated during separate votes by the nearly 1,000 delegates gathered for the United Methodist Church's General Conference in Tampa, Fla. One proposal would have replaced the "incompatible" phrase in the Book of Discipline, which contains the denomination's laws and doctrines. High School Yearbook Advisor Slammed For 'It's OK To Be Gay' Student Profile
A Tennessee high school journalism professor has infuriated locals, including one school board member, after publishing a pro-gay profile of a graduating student in the yearbook. Written by a member of the Lenoir City High School yearbook's student staff, "<a href="http://www.towleroad.com/2012/05/christian-school-board-member-stirs-firestorm-in-tennessee-over-its-okay-to-be-gay-student-yearbook-.html" target="_hplink">It's OK to be Gay</a>" describes the experiences of openly gay student Zac Mitchell. In the piece, Mitchell describes coming out in public and being bullied, as well as cross-dressing and being "hit on by straight guys," <a href="http://www.knoxnews.com/news/2012/may/02/high-school-yearbook-article-sparks-furor-in/" target="_hplink">according to the <em>Knoxville Sentinel</a></em>. Prominent Russian Gay Rights Activist Fined Under 'Propaganda' Law
A prominent Russian gay rights activist was convicted on Friday of spreading "gay propaganda" among minors in the first such ruling in Russia's modern history. Nikolai Alexeyev told the Associated Press that a city court in St. Petersburg fined him 5,000 rubles ($170) for breaching the law, which was controversially introduced by lawmakers in Russia's second-largest city in February. California Transgender Woman Killed In Alleged Hate Crime
Friends, family members and LGBT advocates are mourning the death of a California transgender woman who was gunned down in Oakland over the weekend. <a href="http://abclocal.go.com/kgo/story?section=news/local/east_bay&id=8641958" target="_hplink">As ABC7 is reporting</a>, Brandy Martell was approached by one or two men while sitting behind the wheel of her car early Sunday. Though a witness told the news station the 37-year-old Martell's conversation seemed cordial, one of the men eventually became angry and fired into the car where she was sitting. Reverend Billy Graham Backs Anti-Gay Marriage Amendment In North Carolina
The Rev. Billy Graham urged North Carolina voters Wednesday to support an amendment to the state constitution banning gay marriage, a move that an observer said was highly unusual but another said was in keeping with the minister's moral beliefs. "Watching the moral decline of our country causes me great concern," said Graham, 93, who lives near Asheville. NYC Mayor Bloomberg Plans to Throw Homeless Youths Out of Their Shelter Beds
Yesterday Mayor Bloomberg released a budget that guts a wide array of services to the disadvantaged youths of New York City. But most stunning is his proposal to eliminate 160 of the city's 250 youth shelter beds. How can he possibly think that an appropriate way to deal with budget challenges is to throw poor children out of their shelter beds and into the streets? I wonder if the mayor understands what it means to put a kid on the street. Does he know that many will be forced to resort to prostituting themselves in order to survive? Does he know that 20 percent of the LGBT kids will become infected with HIV on the streets? Does he know that 60 percent will consider or attempt suicide? He ought to know these things, as his administration released a report in 2010 that documented the horrific risks LGBT youths face on the streets. North Carolina Pastor, Slams Gay Marriage, Suggests Unions Between 'Person And A Beast' Could Be Next

Texas Transgender Woman Ticketed For Using Ladies' Room
A Texas-based transgender woman <a href="http://thinkprogress.org/lgbt/2012/05/02/475043/transgender-veteran-bathroom/" target="_hplink">said she was humiliated after being ticketed for using the ladies' room</a> at a Dallas hospital. <a href="http://usnews.msnbc.msn.com/_news/2012/05/02/11501842-transgender-woman-ticketed-for-using-ladies-room?lite" target="_hplink">As MSNBC is reporting</a>, Paula Witherspoon was cited for disorderly conduct on April 25, after a Parkland Hospital official received a complant received a complaint from a female patient about a man in the women's restroom. An officer with the hospital's police force wrote the citation for a class-C misdemeanor. Witherspoon, who has been transitioning since 2006 but lives full-time as a woman, called the experience both "humiliating" and "degrading." She went on to note, "I felt like I was being discriminated against...I went to the bathroom, took care of my business, washed my hands and left.
