(AP) ? Target is setting its bullseye on Canada in 2013.

It's investment in its Canadian launch this year and weaker-than-expected holiday sales caused Target Corp.'s net income to fall 2 percent in the fourth quarter of last year. But the second largest discounter in the U.S. said its foray into Canada, policy of matching competitors' prices and new designer lines will help its business this year.

"We believe that we are well positioned to succeed even in this uncertain environment, said CEO Gregg Steinhafel said in a call with investors.

The big-box retailer, known for its cheap but trendy merchandise, pulled out all the stops to lure in cautious shoppers during the winter holiday season, which runs from November through December. It launched a line of gifts created by 24 high profile designers in partnership with luxury department store Neiman Marcus and offered to match prices of online competitors such as Amazon.com, Walmart.com, Bestbuy.com and Toysrus.com.

But the initiatives did not spur customers to buy more during the period, a time when retailers can make up as much as 40 percent of their annual revenue. In fact, Target noted that sales "bar belled," meaning that they were strong around Black Friday, the busy shopping day after Thanksgiving, and the days before and after Christmas, but not in between.

Overall, Target's number of transactions fell 1 percent during the quarter, although the amount spent per transaction rose 1.4 percent. The company said its gross margin ? the percentage of each dollar in revenue made that a company actually keeps ? declined during the quarter due to holiday markdowns.

Edward Jones analyst Brian Yarbrough said the problem during the holidays was seasonal merchandise that didn't sell during a slow December. But overall it was a decent report, he added.

"Results were in-line, and forward guidance was pretty solid," he said.

Many had wondered how Target would do now that consumers are being squeezed by an increase in the Social Security payroll tax of 2 percentage points that was rolled out last month. Burger King and Wal-Mart have already noticed a pull-back from the tax hike.

Steinhafel did not mention an effect in the fourth quarter, but said that the payroll tax increase was one of several challenges that are giving the company a "tempered" view of revenue in 2013.

"While there are some encouraging signs in the housing market, volatility in consumer confidence, the payroll tax increase and rise in the price of gas all present incremental headwinds," Steinhafel said Wednesday in a call with analysts.

For now, Target, which has 1,778 stores across the U.S., is focusing on its launch in Canada. The company plans to open 24 Canadian stores by early April, one of five waves that should add up to 124 stores open before the end of the year. In addition, the chain plans to open 15 to 20 new stores in the U.S. It is the most new stores the company has opened in one year in its history.

"This is a big investment, people are going to be anxious to see how those are trending," said Yarbrough, the Edward Jones analyst.

For the three months ended Feb. 2, Target earned $961 million, or $1.47 per share, for the period ended Feb. 2. That's down from $981 million, or $1.45 per share, a year earlier.

Removing costs related to its investment in Canada stores and interest expense, earnings were $1.65 per share. Analysts expected earnings of $1.47 per share, on average, according to research firm FactSet, but some may have included the Canadian expense in their projections. Target had forecast adjusted earnings between $1.64 and $1.74 per share.

Revenue climbed 7 percent to $22.73 billion from $21.29 billion. This met Wall Street's expectations.

During the quarter, revenue at stores open at least a year edged up 0.4 percent. This figure is a key indicator of a retailer's health because it excludes results from stores recently opened or closed.

For the full year, the Minneapolis company earned $3 billion, or $4.52 per share. A year earlier it earned $2.93 billion, or $4.28 per share. Adjusted earnings were $4.76 per share. Annual revenue increased 5 percent to $71.96 billion from $68.47 billion.

Target's outlook for 2013 was brighter. The chain foresees first quarter adjusted earnings of $1.10 to $1.20 per share. Analysts predict earnings of $1.05 per share. It expects revenue in stores open at least one year to be flat to up 2 percent.

The chain's fiscal 2013 outlook is for adjusted earnings between $4.85 and $5.05 per share. Wall Street expects earnings of $4.87 per share.

In a call with investors, Chief Financial Officer John Mulligan said he expects revenue in stores open at least one year to be in line with the current year's 2.7 percent rise.

Target's stock fell 65 cents to $63.40.

MADRID, Feb 27 (Reuters) - Lionel Messi has rarely been accused of failing to deliver in big games, having scored in two European Cup finals, but after subdued performances against AC Milan and Real Madrid, questions are being asked. Inmarsat announces new scalable L-TAC service for defence market ...

Inmarsat

Delivers robust, low cost tactical satellite communications from users? existing military radios

MilSatCom, Dubai, 28 February 2013: Inmarsat, (LSE:ISAT.L), the leading provider of global mobile satellite communications services, is expanding the availability of mobile tactical Beyond-Line-of-Sight (BLOS) communications to a broad range of new government users through the innovative exploitation of the capabilities of its L-band satellites.

The company today announced its plan to launch L-TAC, a new L-band service, which will deliver a ?UHF-like? tactical satellite capability for use with existing UHF tactical radios for approved government customers at low cost.

UHF tactical satellite capability is in very high demand by government customers because of its suitability to create Beyond-Line-of-Sight Push-to-Talk networks using tactical radios either man-portable or installed in vehicles, helicopters, ships or other mobile platforms.? Inmarsat?s L-TAC service complements this existing UHF tactical capacity, providing additional capability when UHF capacity is unavailable. Inmarsat?s powerful Inmarsat-4 constellation of satellites provides this capability across the world, with the additional benefit of supporting small antennas for BLOS communications on the move.

To access the service, users require only a small antenna adaptor for their existing tactical radio, which replaces their existing UHF antenna.? Inmarsat has partnered with Spectra Group to develop the Slingshot? adaptor, which is expected to be commercially available in late Q2 2013.? Designed with the soldier in mind, Slingshot? will work with existing tactical military radios and with minimal additional training, will provide Beyond-Line-of-Sight communications without the need for additional infrastructure. Slingshot? combined with Inmarsat?s L-TAC leased service is fully flexible and designed to meet security and reliability requirements cost-effectively. ?Users will be able to lease the service for a fixed period with leases as short as one month.

Andy Start, President, Inmarsat Global Government, said: ?We are delighted to be launching this new service in partnership with Spectra. L-TAC will enable militaries to exercise greater command and control through existing tactical radios at a realistic cost with leases available for periods as short as one month at a time. The operational benefits for soldiers on the ground are enormous, providing low risk, highly resilient and easy to use connectivity independent of local infrastructure or terrain.?

Simon Davies, Managing Director, Spectra Group commented: ?This service will enable governments to provide enhanced satellite and radio communications without large capital expenditure. As governments around the world review military budgets, this is a highly compelling offer.?

Azerbaijanis injured in fight at French parliament

PARIS (AP) ? French officials on Thursday condemned Armenian and Azerbaijani activists for clashing with one another in France's parliament.

Azerbaijan and Armenia have been in conflict since 1988, over the disputed Caucasus Mountains territory of Nagorno-Karabakh.

The violent outburst occurred Tuesday in the National Assembly in Paris, during a conference commemorating the anniversary of violence against Armenians in 1988.

Two Azerbaijanis, a man and a woman, tried to interrupt the conference, saying they wanted to commemorate the anniversary of a 1992 massacre of Azerbaijanis. The Azerbaijani Embassy in Paris says the two were attacked and beaten by some of the pro-Armenia participants.

France's Foreign Ministry said Thursday it "regrets and condemns" the acts of violence committed inside the National Assembly.

The Azerbaijani Foreign Ministry issued a statement criticizing French lawmakers for failing to stop the "brutal beating" of Azerbaijani activists. It blamed "the Armenian lobby and its sponsors" for the violence and voiced hope that French authorities will punish those responsible for the beating the woman, a French citizen of Azerbaijani origin, and an Azerbaijani student.

In Azerbaijan's capital, Baku, about 100 student activists protested near the French Embassy, holding placards with slogans such as "Shame on France!" and "For Justice!"

3D? Feh. MIT has already moved on to 4D printing (video)

3D Feh MIT has already moved on to 4D printing video

The bad news: just as much of the world is starting to get excited about the prospects of 3D printing, science is moving on to the world of 4D. The good news: in the future, you might not have to assemble that Ikea chair yourself. "4D printing" is the term cientists are using to refer to a technology that MIT's Skylar Tibbits talked up during a recent TED appearance. The fourth "d" here is time, referring to an object that, once printed, is capable of changing shape (over time, naturally).

"Essentially the printing is nothing new," Tibbits told the BBC. "It is about what happens after." So far the concept has been demonstrated with thin strands of plastic, which, once added to water, form into a predetermined shape, using energy from the absorption. Suggested future applications involve furniture, pipes, bikes and buildings. First, however, scientists will have to demonstrate the technology on a larger structure, of course, and they'll explore the possibility of other energy sources, like heat, sound and vibration.

'Top Chef' finale: Will Kristen or Brooke win?

David Moir/Bravo

By Drusilla Moorhouse, TODAY contributor

For "Top Chef: Seattle" finalists Kristen Kish and Brooke Williamson, it's time to unpack their knives and go!

In Wednesday night's season 10 finale, the ladies will prepare five of their best dishes -- live in front of a studio audience. In addition to their own guests, the spectators will include the champions of all nine previous seasons of "Top Chef."

(In other words, no need to fear a repeat of season nine's cross-country ski/target-shooting "Culinary Olympics." What that had to do with cooking, we're not sure.)

Brooke, the steely owner of two acclaimed Los Angeles gastropubs, has dominated the competition for weeks. Then again, she hasn't had to square off against Kristen since the former model was eliminated in the 11th round.

After winning four challenges, Kristen was sent home during the infamous Restaurant Wars. (While Josie stayed. Boo! Hiss!) But then she sliced and diced five other eliminated cheftestants in the Last Chance Kitchen to earn her spot in the finale.

Who do you hope will win the title of Top Chef? Sound off on our Facebook page!

Managing Stress As a Business Manager : American Business ...

StressConsulting experience and various international studies have shown that business managers in all types and sizes of companies have very difficult and stressful jobs. In today?s tough, dynamic, resource-tight and uncertain economy, a company (or organization) needs strong managers to lead its staff toward accomplishing business goals.

But managers are not only leaders. They are problem solvers, coordinators, communicators, cheerleaders and planners as well. And managers don?t come in onesize- fits-all shapes or forms. Managers fulfill many roles and have many different responsibilities at each level of management within an organization.

Organizations abound in today?s society. Groups of individuals constantly join forces to accomplish common goals. Sometimes the goals of these organizations are for profit, such as franchise restaurant chains or clothing retailers. Other times, the goals are more altruistic, such as nonprofit churches or public schools.

But no matter what their aims, they?re made up of people and certain individuals are in charge of these people. These are the managers.

Managers appear in every organization because organizations want to succeed. These individuals have the sometimes unenviable task of making decisions, solving difficult problems, setting goals, planning strategies and rallying individuals. And those are just a few of their responsibilities.

To be exact, managers manage themselves, administer and coordinate resources effectively and efficiently to achieve the goals of an organization, manage context, manage relationships and manage change. In essence, managers get the job done effectively, especially if they manage both themselves and other people very well.

No matter what type of organization they work in, managers are generally responsible for the performance of a group of people. As leaders, managers must encourage this group to reach common organizational goals (at the general level), such as bringing a new product to market in a timely fashion and also reach specific objectives, such as improve profits by a certain percentage by the end of a given time period.

To accomplish these general goals and specific objectives, in an organizational products and services framework, managers not only use their human resources, but they also use inputs, methods and systems, to create outputs which can be translated into products and services for customers and a control system to achieve all these in the most optimal way.

When you take into consideration the full array of duties, roles and responsibilities in leading and managing their units, departments, organizations, etc., one thing becomes clear: business managers and professionals have difficult jobs. Huge Protest Vote Leaves Italy Facing Deadlock

* Global markets shaken by fears of new euro zone instability
* Vote is stunning success for populist movement
* Berlusconi stages comeback but cannot govern alone
* Centre-left big losers although will try to form government
By Barry Moody and James Mackenzie
ROME, Feb 25 (Reuters) - A huge protest vote by Italians enraged by economic hardship and political corruption left the euro zone's third-largest economy facing a dangerous vacuum on Monday after an election in which no group won enough votes to form a government.
The result, in which anti-euro parties took more than 50 percent of the vote and a novice populist movement scored a stunning success, rocked global markets with fears of a new euro zone crisis.
Europe's common currency slumped against the dollar and yen and U.S. stocks suffered their biggest one-day drop since November.
With more than 99 percent of returns in from polling stations, results showed the centre-left had taken a slim victory of around 130,000 votes in the lower house of parliament, enough to give it comfortable control thanks to a big winner's bonus.
But no party or likely coalition won enough seats to form a majority in the upper house, creating a deadlocked parliament - the opposite of the stable result that Italy desperately needs to tackle a deep recession, rising unemployment and a massive public debt.
The outcome fanned fears of a new European financial crisis, with prospects of a long period of paralysis and uncertainty in Italy.
"This is the worst possible outcome from the market's point of view ... It seems inevitable that there will be a new election," said Alessandro Tentori, Citigroup head of global rates.
The result was an extraordinary success for Genoese comic Beppe Grillo, leader of the populist 5-Star Movement, who toured the country in his first national election campaign hurling obscenity-laced insults against a discredited political class.
He was set to become the biggest single party in the lower house, riding a potent wave of anger against rampant waste and corruption by ageing political leaders.
His success fulfilled the predictions of some analysts that the most uncertain and closely watched election in years would herald a political revolution. "This is the end of a system, not a government," respected commentator Massimo Franco told Reuters before the vote.
Grillo polled more around a quarter of the vote in a meteoric rise from the 1.8 percent he garnered in his movement's first local political test in 2010.
The result was a humiliating slap in the face for colourless centre-left leader Pier Luigi Bersani, who threw away a 10-point opinion-poll lead less than two months ago against Silvio Berlusconi's centre right.
He failed to turn up for a press conference after the result became clear. His deputy, Enrico Letta, as well as outgoing technocrat premier Mario Monti, said responsible forces must form a government and avoid another election. But the result raised a big question over whether that would be possible.
Billionaire media magnate Berlusconi, 76, who staged an extraordinary comeback from sex and corruption scandals since diving into the campaign in December, came in a close second in the Senate race, with an estimated 117 seats.
With almost all results in, the centre-left was set to take 121 seats in the upper house, Grillo 54, and Monti languishing on only 22 after a campaign which never took off. The Senate majority is 158.
Berlusconi, a master politician and communicator, wooed voters with a blitz of television appearances and promises to refund Monti's hated housing tax despite accusations from opponents that this was an impossible vote buying trick.
Grillo has attacked all sides in the campaign and ruled out a formal alliance with any group although it was not immediately known how he would react to his stunning success or how his supporters would behave in parliament.
The next move to solve the crisis will be when head of state Giorgio Napolitano calls in political leaders to discuss how to form a government. But this is not expected until March 10 after the election result is formally confirmed and parliament convened.
Letta said the centre-left, as biggest party in the lower house, had the right to be the first to try to form a government.

DANGER OF NEW ELECTION
Investors fear a return of the kind of debt crisis that took the euro zone close to disaster and brought the technocrat Monti to office, replacing Berlusconi, in 2011.
The results showed more than half of Italians had voted for the anti-euro platforms of Berlusconi and Grillo.
A centre-left government either alone or ruling with Monti had been seen by investors as the best guarantee of measures to combat a deep recession and stagnant growth in Italy, which is pivotal to stability in the currency union.
But the failure of Monti to gain traction at the head of a centrist force, despite support from business leaders and foreign governments, and the weak showing by the centre-left meant they do not have nearly enough Senators to do this.
The upper and lower houses have equal law-making power.
The benchmark spread between Italian 10-year bonds and their German equivalent widened from below 260 basis points to above 300 and the Italian share index lost all its previous gains after projections of the Senate result.

Monti helped save Italy from a debt crisis when Rome's borrowing costs were spiralling out of control in November 2011, but few Italians now see him as the saviour of the country, which is reeling under its longest recession for 20 years.
Grillo's movement rode a wave of voter anger about both the pain of Monti's austerity programme and a string of political and corporate scandals. It had particular appeal for a frustrated younger generation shut out of full-time jobs.
"I'm sick of the scandals and the stealing," said Paolo Gentile, a 49-year-old Rome lawyer who voted for 5-Star. "We need some young, new people in parliament, not the old parties that are totally discredited."
Berlusconi, a billionaire media tycoon, exploited anger against Monti's austerity programme, accusing him of being a puppet of German Chancellor Angela Merkel, but in many areas Grillo was a bigger beneficiary of public discontent.
Italians wrung their hands at prospects of an inconclusive result that will mean more delays to essential reforms.
"It's a classic result. Typically Italian. It means the country is not united. It is an expression of a country that does not work. I knew this would happen," said 36-year-old Rome office worker Roberta Federica.
Another office worker, Elisabetta Carlotta, 46, shook her head in disbelief. "We can't go on like this," she said.

(AP) ? A ranking BP executive testified Tuesday that the London-based oil giant and its contractors share the responsibility for preventing blowouts like the one that killed 11 workers and spawned the nation's worst offshore oil spill in 2010.

Lamar McKay, who was president of BP America at the time of the Deepwater Horizon disaster, became the first BP executive to testify at a federal trial intended to identify the causes of BP's Macondo well blowout and assign percentages of blame to the companies involved.

Rig owner Transocean and cement contractor Halliburton also are defendants at trial, which opened Monday.

A plaintiffs' attorney pressed McKay to agree with him that BP bore ultimate responsibility for the blowout, but McKay insisted that managing the hazards of deepwater drilling are a "team effort."

"I think that's a shared responsibility, to manage the safety and the risk," said McKay, now chief executive of BP's Upstream unit. "Sometimes contractors manage that risk. Sometimes we do. Most of the time it's a team effort."

McKay also defended BP's internal probe of the spill, which outlined a series of mistakes by rig workers and faulted decisions by other companies but didn't assign any blame to BP's upper-level management.

"I think it was a substantial investigation," McKay said. "I think we've learned what we can from the accident and we're trying to put those things into practice right now."

McKay, whose testimony will resume Wednesday, called the disaster a "tragic accident" resulting from a "risk that was identified."

It wasn't the first time McKay testified under oath about the spill. He appeared before Congress less than a month after the explosion.

U.S. District Judge Carl Barbier is hearing the case without a jury. Barring a settlement, Barbier will decide how much more money BP and other companies owe for their roles in the disaster.

McKay's testimony followed that of an expert witness for people and businesses suing the company. University of California-Berkeley engineering professor Robert Bea testified that BP PLC didn't implement a 2-year-old safety management program on the rig, which exploded on April 20, 2010.

"It's a classic failure of management and leadership in BP," said Bea, a former BP consultant who also investigated the 1989 Exxon Valdez spill and New Orleans levee breaches after Hurricane Katrina in 2005.

BP has said its "Operating Management System" was designed to drive a rigorous and systematic approach to safety and risk management. During cross-examination by a BP lawyer, Bea said the company made "significant efforts" to improve safety management as early as 2003.

However, the plaintiffs say BP only implemented its new safety plan at just one of the seven rigs the company owned or leased in the Gulf at the time of the disaster.

Bea said it was "tragic" and "egregious" that BP didn't apply its own safety program to the Deepwater Horizon before the Macondo well blowout. Transocean owned the rig; BP leased it.

BP lawyer Mike Brock said the company allows contractors like Transocean to take the primary responsibility for the safety of rig operations as long as the contractor's safety system is compatible with BP's ? an arrangement that Brock suggested is a standard industry practice.

In his May 2010 congressional testimony, McKay said BP's Operating Management System is "as good as anyone."

"I know of nothing that points me in a direction that we have deficiencies" in the system, McKay said.

As he questioned Bea, Brock recited a long list of steps that BP took to improve safety, citing them as evidence that the company wasn't "cutting corners" on safety.

A plaintiffs' lawyer showed Bea a transcript of a deposition of Tony Hayward, BP's CEO at the time of the disaster. Hayward was asked if the blowout could have been averted if BP had implemented the safety management program in the Gulf.

"There is possible potential," Hayward responded. "Undoubtedly."

Bea said BP's "culture of every dollar counts" was reflected in a May 2009 email sent by BP well team leader John Guide: "The DW Horizon embraced every dollar matters since I arrived 18 months ago," Guide wrote. "We have saved BP millions and no one had to tell us."

"Financially, BP had the resources to effectively put into place a process safety system that could have prevented the Macondo disaster," Bea testified.

BP has already pleaded guilty to manslaughter and other criminal charges and has racked up more than $24 billion in spill-related expenses, including cleanup costs, compensation for businesses and individuals, and $4 billion in criminal penalties.

Plaintiffs' attorney Robert Cunningham read portions of the plea agreement as he pressed McKay to say how much responsibility BP takes for the catastrophe. Cunningham noted that nothing in the document assigns blame to specific BP executives.

"That is not written in there. That's true," McKay said.

Two BP rig supervisors, however, have been indicted on manslaughter charges for the workers' deaths and are awaiting a separate trial.

"There were some misinterpretations and mistakes made" on the rig, McKay said.

One of the biggest questions facing Barbier is whether BP acted with gross negligence.

Under the Clean Water Act, a polluter can be forced to pay a minimum of $1,100 per barrel of spilled oil; the fines nearly quadruple to about $4,300 a barrel for companies found grossly negligent, meaning BP could be on the hook for nearly $18 billion.

90% No

All Critics (58) | Top Critics (16) | Fresh (52) | Rotten (6)

Nominated by the Academy as the year's best foreign-language film, No grabs you hard, no mercy, and keeps you riveted.

Larra?n's unarguable point is that, in politics, if we wait for good to issue only from the pure in heart, we will be waiting a very long time.

[Lorrain has] made a few daring choices here, not all of which work.

A troubling, exhilarating and ingeniously realized film that's part stirring political drama and part devilish media satire ...

For anyone fascinated by the political process and the powers of persuasive advertising, No is a resounding yes.

It hangs on three ideas...While each...is intriguing, the execution of all is less than satisfying.

Larra?n's script is punctuated by dark bursts of humour, and the filmmaker knowingly navigates his audience to a nail-biting - though never cloying, and fully warranted - climax.

It makes the superficial Mad Men seem like, well, a commercial. Buy, buy, buy.

A fascinating period re-creation if not an especially compelling drama.

Evocative and suspenseful, the film is a fascinating glimpse into recent history and the democratic process.

The film highlights the sad fact that logical arguments don't win political debates or elections. Sloganeering and advertising do.

Using a technique borrowed from cinema verit? documentaries, the director succeeds in making us feel as if we're living each moment right alongside his politically-charged characters.

It's a perfectly fine movie, but given its fairly radical storyline, the filmmaking tends to hew toward the safe and the familiar.

"No" gives a fresh look at the little known history of a country whose duly elected government under Salvador Allende was overthrown in a military coup led by Pinochet in 1973.

Savvy, often brilliant ...

Bernal plays the creative type perfectly. His big eyes always seem to be seeing things that others don't, and through his calm, methodical demeanor, you can sense the wheels turning in his head.

Donkey, buffalo found in South African meat products

CAPE TOWN (Reuters) - Donkey, water buffalo and other unconventional ingredients have been found in almost two thirds of hamburgers and sausages tested in South Africa, a study released on Tuesday showed.

The tests by the University of Stellenbosch were planned before a scandal broke out in Europe over horsemeat labelled as beef that raised concerns worldwide over the risks to human health from a complex and nebulous meat supply chain.

"Our study confirms that the mislabelling of processed meats is commonplace in South Africa and not only violates food labelling regulations, but also poses economic, religious, ethical and health impacts," co-author Louw Hoffman of the university's Department of Animal Sciences, said in a statement.

Soya, donkey, goat, water buffalo and plant material were found in up to 68 percent of the 139 minced meats, burger patties, delicatessen meats, sausages and dried meats tested by the university. The items were not listed as ingredients.

Pork and chicken were the most common fillers found in products that were not supposed to contain them, according to the study that used DNA testing techniques and was published in the journal Food Control.

No similar discoveries had been made over the past two years, when DNA testing became more widely used in South Africa.

Stricter food labelling laws came into effect in the continent's largest economy in March last year, with mandatory information required on content, country of origin and allergens.

But there is no mandatory government testing of food sold in South Africa.

"It is a wake-up call for the industry to abide with the new labelling regulations," Hoffman, a noted game meat researcher, told Reuters.

He said extensive tests over two weeks of more than 100 samples had found no trace of horse meat.

Scientists' findings disclose a new and much needed test for river blindness infection

Scientists' findings disclose a new and much needed test for river blindness infection

Tuesday, February 26, 2013

Scientists at The Scripps Research Institute (TSRI) have found a telltale molecular marker for Onchocerciasis or "river blindness," a parasitic infection that affects tens of millions of people in Africa, Latin America and other tropical regions. The newly discovered biomarker, detectable in patients' urine, is secreted by Onchocerca volvulus worms during an active infection. The biomarker could form the basis of a portable, field-ready test with significant advantages over current diagnostic methods.

"There has been a need for an inexpensive, non-invasive test that can discriminate between active and non-active river blindness infections during treatment campaigns," said Kim D. Janda, who is Professor and Ely R. Callaway, Jr. Chair in Chemistry, member of the Skaggs Institute for Chemical Biology, and director of the Worm Institute of Research and Medicine at TSRI. "We think that this new biomarker can be the basis for such a test."

The work is described in an online Early Edition of the Proceedings of the National Academy of Sciences during the week of February 25, 2013.

Leading Cause of Vision Loss

A leading cause of vision loss, Onchocerciasis infections are transmitted among humans by river-dwelling blackflies in tropical regions. The vast majority of cases occur in sub-Saharan Africa, although pockets of endemic infection exist in Yemen and in Central and South America. The major symptoms of the disease, including blindness, result from the spread of O. volvulus "microfilariae"?early-stage larval worms?to the eyes and other tissues, where they trigger damaging inflammatory reactions.

Mass treatment campaigns, begun in the 1990s, have used the anti-worm drug ivermectin, as well as the antibiotic doxycycline, which kills a symbiotic bacterium within the worms. The World Health Organization's African Programme for Onchocerciasis Control has set a target date of 2025 for the eradication of the disease in that region. But Onchocerciasis treatment is seldom effective immediately and often spares adult worms. The latter can remain in protected nodules under the skin of a patient and secrete microfilaria for a decade or more. Health agencies need better diagnostic methods not only to monitor the progress of Onchocerciasis treatment campaigns, but also to limit the use of ivermectin and doxycycline to reduce the risk of resistance.

Current diagnostic methods include the painful cutting of "skin snips" from patients for microscopic analysis and an ELISA antibody test for microfilariae, which may yield positive results even for non-active infections. "You can still have circulating antibodies to a nemotode antigen in your blood for a long time after the infection is gone," said Janda.

Looking for a Better Way

A better diagnostic marker would be a metabolite of O. volvulus that appears only during an active, microfilariae-producing infection and that could determine both the presence and the severity of disease. In 2010, Janda's laboratory demonstrated the feasibility of this approach by sifting through the small-molecule metabolites within blood samples from river blindness patients?a technique called "metabolome mining"?and finding a set linked to active onchocerciasis infection. For the new study, the team sought a simpler set of biomarkers?or better yet, a single unique biomarker in urine.

Daniel Globisch, a postdoctoral fellow in the Janda laboratory, started with samples of urine from onchocerciasis-infected and non-infected Africans. Using a powerful laboratory technique called liquid chromatography mass spectrometry, he measured the concentrations of hundreds of small-molecule metabolites in the samples. Excitingly, between the infected and non-infected urine samples, one difference stood out clearly: "An unknown small molecule was highly elevated in the samples from infected individuals," said Globisch.

In a process akin to looking for the proverbial needle in the haystack, Globisch was able to purify the mysterious metabolite, and, using mass spectrometry, determine the chemical identities of its individual pieces. "The metabolite itself wasn't present in the databases, so I searched the literature for what is known about the biosynthesis and metabolic pathways in these nematodes," Globisch said. Ultimately, he was able to identify the metabolite as N-acetyltyramine-O,?-glucuronide. Remarkably, this molecule's inception can be traced to O. volvulus as a neurotransmitter molecule that is secreted by young, reproducing worms and then modified by the human body on its way to being excreted in urine.

"It's a spectacular find in terms of biomarkers as it does not occur naturally in humans," Globisch said. Levels of the metabolite in a non-infected North American control sample were near zero.

Toward a Field Test

In urine samples from Africans with active onchocerciasis infections, Globisch found that levels of the biomarker were on average four to six times higher than in samples from Africans with non-active infections. In a separate test, the team determined that a full course of doxycycline treatment, which sterilizes or kills infecting worms by destroying their symbiotic bacteria, also reduced levels of the biomarker to near-normal. "This biomarker appears to be specific for an active infection," Globisch said. The wide gap between biomarker levels in active and non-active infections suggests that a field test based on the biomarker would be robustly useful.

Such a diagnostic, said Janda, might ultimately be a simple urine dipstick test, much like a home pregnancy test, which would indicate the amount of the O. volvulus biomarker present in the sample. "Ultimately for this to be of value in Third World countries we will need to morph this biomarker into something that's inexpensive, simple to use, tolerant of extreme temperatures and portable?basically distilling our finding to a test that can be carted around in a backpack," Janda said.

Importantly, he adds that Globisch's metabolome-mining approach in theory should be applicable to the development of diagnostic tests for other worm diseases.

First Person: Forever Changed After Service in Iraq

One of the last photos taken of me before I left the service in 2011. As we near the 10-year anniversary of the start of the war in Iraq, Yahoo News asked U.S. servicemen and women who served to share their perspectives and discuss how it changed them. Here's one story.

FIRST PERSON | On March 19, 2003, when the U.S. military invaded Iraq, I was 18 and in Navy basic training. We had no TV, but the recruits were taken elsewhere to watch the televised event unfold. I didn't go because I was standing watch, and my training commander said I would miss the fun. I responded that I would most likely witness it first-hand myself one day.

I deployed to Iraq twice. The first was in 2005 aboard the USS Carl Vinson, and the second was in 2007-2008, embedded with the Army in Baghdad, Iraq and it's this deployment I'd like to discuss.

I was 23 and a Second Class Petty Officer (E-5). My task was to document the war for historical preservation. I traveled the country to collect data, pictures, artifacts and I interviewed multiple individuals to ensure the efforts of our military would be remembered.

A Marine colonel pins the Iraq Campaign Medal as well as a Joint Service Commendation Medal to my uniform.I witnessed and learned of many acts that ranged from heroic to atrocious. I interviewed troops who described events endured like the loss of comrades, close calls in combat and the overall daily life of service in Iraq.

Many Americans are at least aware of the videos terrorists posted online of beheadings and other atrocities, but because of my job I saw hundreds. One particular video was of a man who cried his children's names while begging his captors for his life. He had three daughters, and I watched this video as he was violently executed. It's now a rare occurrence, but I occasionally wake up hearing his screams echoing in my head.

I can't answer what we as a nation gained by invading Iraq, or whether invading was the right thing to do. What I can say is my belief as to why our service members answer our nation's call, regardless of where it takes them or the reasoning. I believe troops fight not for love of country nor orders handed down, but because of those who stand beside them. They fight because the men and women one serves with become his family, and these warriors selflessly risk their own lives for those of the family who surround them in arms.

A picture taken outside a checkpoint near a base in Baghdad, Iraq.Iraq affected my life in many ways. I was diagnosed with PTSD in 2008 and it wasn't caused by the close calls on my life, but the atrocities I was made aware of and my inability to come to terms with them. This cost me my first true relationship, pushed me to the brink of contemplating suicide and required therapy to learn to live with my knowledge of human depravity.

I am now a forever changed, but happy man. Iraq will always be with me, but I know how to cope and encourage anyone suffering symptoms of PTSD to seek help.

If asked to do it again, even if it meant sacrificing everything, I would without hesitation. I would because standing shoulder-to-shoulder with such high-caliber warriors, at sea and in Iraq, was the most rewarding experience of my life.

Sterling struggles after UK debt downgrade

LONDON (AP) ? The British pound is recovering after dropping against the world's leading currencies as markets reacted to a downgrade of the U.K.'s cherished triple-A credit rating.

During early trading Monday, sterling dropped to $1.5069 against the dollar ? its lowest level since July 2010 ? before bouncing back to $1.5144. Against the euro, the pound hovered around 18-month lows with one euro worth 0.8745 pound.

The pound was in focus in the wake of last Friday's decision by Moody's to downgrade the U.K.'s credit rating by one notch from the top AAA to AA1. The agency says sluggish growth and rising debt are weakening the British economy's outlook.

Two other major rating agencies ? Fitch and Standard & Poor's ? have also warned that they may downgrade the U.K. as well.

Seth MacFarlane Wasn't a Terrible Oscars Host After All

Minutes into his opening monologue as host of the 2013 Oscars, Seth MacFarlane cracked a joke about the best oicture nominee, Django Unchained. "This is the story of a man fighting to get back his woman, who's been subjected to unthinkable violence," he began. "Or, as Chris Brown and Rihanna call it, a date movie." While the audience laughed uneasily, MacFarlane voiced what many were thinking about the host: "'That's what we were afraid he would do,'" he quipped. "That's as bad as it gets, if it makes you feel any better!" he tried to assure viewers (and censors).

TSX slumps, hit by fears of Italian gridlock

TORONTO (Reuters) - The prospect of a divided Italian parliament prompted a late retreat in banking and other Canadian financial stocks on Monday, pushing the main Toronto equity index into the red after it had earlier hit a three-week high.

Voting projections in Italy show no coalition garnered enough votes to form a government, reviving fears about an extended period of uncertainty in Europe's third-largest economy.

"The basic problem is that the whole Europe situation had been shunted to the back-burner ever since the beginning of this year," said Elvis Picardo, strategist at Global Securities in Vancouver.

"The latest developments show that there is a degree of risk that is still attached to the continent," he said. "It's causing investors to rethink some of their assumptions about the market."

Royal Bank of Canada fell 1 percent to C$63.59, insurer Manulife Financial Corp lost 2.1 percent to C$14.84 and its rival, Sunlife Financial Inc , slipped 1.9 percent to C$28.34.

Picardo said investors may also be feeling more cautious about bank earnings, which are due to be reported this week and next, after a recent string of weak domestic housing, employment, inflation and retail sales data. <.to/>

The Toronto Stock Exchange's S&P/TSX composite index <.gsptse> ended down 50.76 points, or 0.40 percent, at 12,650.87.

It had earlier hit 12,832.71, its highest level since January 30.

The tide turned in the last hour of trade, and seven of the 10 sectors ended in negative territory.

Energy stocks took a beating, with Suncor Energy Inc falling 1.4 percent to C$31.52 to end the day as the heaviest weight on the index.

It had earlier risen, along with fellow oil sands producer Canadian Natural Resources Ltd , after a Barron's article suggesting each stock could gain 25 percent in the next year.

Canadian Natural Resources finished up 0.6 percent at C30.55.

Gold miners provided some of the only gains, as the price of bullion rose in part on the back of the Italian uncertainty.

Barrick Gold, the world's biggest producer, gained 2.2 percent to C$31.81, while four more gold miners rounded out the top five positive influences on the index.

Shares in BlackBerry ended flat at C$13.48, giving up early gains after the smartphone maker's chief executive told a German newspaper that sales of its make-or-break BB10 line were going better than expected and that the company had increased production to keep up.

Stern calls Clive Davis comments 'sickening'

Louis Lanzano / AP file

By Us Weekly

Howard Stern is adding his voice to the very public, very messy back-and-forth between Kelly Clarkson and Clive Davis. The radio shock jock, who is known more often for slamming celebrities than defending them, took a surprisingly sympathetic stance toward the "American Idol" alum's accusations that Davis had "bullied" her during their collaborations -- and mischaracterized her in his memoir, "The Soundtrack of My Life."

"I always find it sickening when management guys like to set the record straight about how f--king creative and what geniuses they are," Stern said on his show on Wednesday, Feb. 20. "And, like, this guy's trying to diminish what she does? Doesn't he have enough in life? Can't he sort of tell his story without f--king degrading her and putting her down?"

PHOTOS: Shocking celeb feuds

Earlier this week, Clarkson, 30, took to her blog to slam what she called Davis' "memory lapses and misinformation."

"First, he says I burst into 'hysterical sobbing' in his office when he demanded 'Since U Been Gone' be on my album. Not true at all," she wrote of her 2004 album?"Breakaway." "I cried because he hated ["Because of You"] and told me verbatim that I was a 's--tty writer who should be grateful for the gifts that he bestows upon me.'"?

The legendary 80-year-old record producer defended the details of his memoir, though he did own up to "a few creative differences' in their years working together.

PHOTOS: Kelly's funny night at the Grammys 2013

"As anyone who has read 'The Soundtrack of My Life' knows, I think Kelly Clarkson is a tremendous vocal talent and performer," he wrote on Twitter one day later, on Wednesday, Feb. 20. "In the book, I provide an in-depth look at our years together during which we shared major multi-platinum success, as well as a few creative differences. I am truly very sorry that she has decided to take issue with what I know to be an accurate depiction of our time together."

Stern's comments Wednesday similarly praised Clarkson's vocal prowess, though he too saw Davis' memoir as a knock on the "Stronger" singer's talents.

PHOTOS: Kelly's body evolution

"I think she has an amazing voice, and the girl has worked hard to get to where she is," he said. "I don't understand Clive Davis' motivation for knocking this chick down."

The 59-year-old radio host is no stranger to controversy himself, having come under fire earlier this year after calling 'Girls' creator Lena Dunham a "little fat girl" and likening the HBO show's sex scenes to "rape" -- he offered Dunham an on-air apology a few days later.

"It makes me feel bad, and I think she is getting the impression that I somehow think she's just a talentless little fat chick," he said.

LG Optimus L3 II hands-on: Jelly Bean comes in a pebble (video)

LG Optimus L3 II handson Jelly Bean comes in a pebble video

The blink-and-you-miss-it phone you see above is the LG Optimus L3 II, the smallest in the Korean manufacturer's trio of style-driven devices. Much like its predecessor, the L3 II is a 3.2-inch Android handset; the difference this time is the fact that it's running Jelly Bean, a firmware version that way too many larger smartphones -- even some new ones -- are still lacking. The twist here is that only Android fans with small hands and no need for raw processing power need apply, as there's only a Qualcomm Snapdragon S1 chipset (MSM7225) and 512MB RAM running the magic behind the show. Additionally, we were greeted by a QVGA (320 x 240) resolution, 3.15MP rear camera and 1,540mAh battery.

The fact that such a small phone with rather "budget-friendly" specs can run Jelly Bean without too much concern is a fact-check to manufacturers that claim their older devices can't be upgraded to it due to fears that it won't perform properly. Granted, the device was slower than we're accustomed to seeing on other Android 4.1 phones, but we have a feeling that it wouldn't be that much different a story if it were using Ice Cream Sandwich.

When it comes to the fit and feel of the L3 II, you probably won't be terribly surprised to learn that it wasn't terribly comfortable, though admittedly we're now conditioned to do hands-ons with phones as large as 5.5-inch (and even 6.1-inch). That said, its pebble-like form factor nearly got buried in our hands and it was difficult to see even the most trivial of apps, thanks to the vastly limited screen real estate. Still, we recognize that this particular size is designed to fit a very specific demographic, and it will likely delight anyone who is in the market for a smaller handset. Regardless of its size, the L3 II at least feels as if it's made with solid build quality. The white version offers a matte finish, while the black remains glossy -- and yes, a massively annoying fingerprint magnet.
